Western blot analysis was performed on whole cell extracts (30 µg lysate) of HeLa (Lane 1), MCF-7 (Lane 2), HEK-293 (Lane 3), Hep G2 (Lane 4), U-2 OS (Lane 5), A549 (Lane 6), SH-SY5Y (Lane 7) and A431 (Lane 8). G-rich sequence factor 1 (GRSF1) is a regulator of post-transcriptional mitochondrial gene expression. It required for assembly of the mitochondrial ribosome and for recruitment of mRNS and IncRNA. GRSF1 binds RNAs containing the 14 base G-rich element. It preferentially binds RNAs transcribed from three contiguous genes on the light strand of mtDNA, the ND6 mRNA, and the long non-coding RNAs for MT-CYB and MT-NDS - each of which contains multiple consensus binding sequences.
