IKK alpha Antibody in Western Blot (WB) — Western blot analysis of CHUK was performed with 10 µg of A549 cells transfected with Transfection Reagent alone (Lane 1), 100nM Non-Targeting control siRNA (Lane 2), or 100nM siRNA against CHUK (Lane 3). Proteins were resolved using a NuPAGE® Novex 4-12% Bis-Tris Gel (Product # NP0322BOX), XCell SureLock™ Electrophoresis System (Product # EI0002), and a protein size ladder. Proteins were wet transferred to a Pierce Nitrocellulose Membrane (Product # 88025) OR Pierce PVDF Membrane (Product # 88518) and blocked with Pierce Starting Block T20 (PBS) Blocking Buffer (Product # 37539) for 1 hour at room temperature. CHUK was detected at ~ 85 kDa using CHUK Rabbit polyclonal antibody (Product # PA5-17803) diluted in Pierce Starting Block T20 (PBS) Blocking Buffer 4°C overnight on a rocking platform. Pierce Goat Anti-Rabbit (Product # 31461) HRP-Conjugated Antibodies at a 1:2500 dilution were used and chemiluminescent detection was performed using Pierce Supersignal West Dura Maximum Sensitivity Substrate (Product # 37071). Relative density of the bands normalized to GAPDH (36 kDa). CHUK Antibody (Product # PA5-17803) confirms silencing of CHUK expression.

IKK alpha Antibody — Antibody specificity was demonstrated by CRISPR-Cas9 mediated knockout of target protein. A loss of signal was observed for target protein in IKK alpha knockout (KO) cell line compared to control cell line using anti- IKK alpha Rabbit Polyclonal antibody (Product # PA5-17803).

Inhibitor of Nuclear factor Kappa-B kinase subunit alpha (IKK alpha) is encoded by the CHUK gene and located on chromosome 10 in humans. It is also known as IKK1 or CHUK (Conserved helix-loop-helix ubiquitous kinase). IKK alpha belongs to the family of serine threonine protein kinase and forms a part of IKK complex. The IKK complex is required for the activation of NF-kB with IKK alpha specifically known to be involved in the non-canocial pathway for NF-kB activation. IKK alpha along with IKK beta and a regulatory protein NEMO (IKK gamma) phosphorylates IkB proteins which targets the later for ubiquitin mediated degradation. It is involved with several cellular processes with extensive studies on its role in inflammation and cell survival.
