Western blot was performed using Anti-IRAK-M Polyclonal Antibody (Product # PA1-30775) and a 60 kDa band corresponding to Interleukin-1 receptor-associated kinase 3 was observed in THP-1 cells differentiated with PMA and not in undifferentiated THP-1 cells. IRAK3 serine/threonine kinase or interleukin-1 receptor-associated kinase 3 is a member of the interleukin-1 receptor-associated kinase protein family which are essential components of the Toll/IL-R immune signal transduction pathways. IRAK3 is primarily expressed in monocytes and macrophages and functions as a negative regulator of Toll-like receptor signaling. IRAK3 mediates critical aspects of innate immunity that result in an immunocompromised state during sepsis. Mutations in IRAK3 are associated with a susceptibility to asthma.
