The K-Ras gene, specifically the G12D mutation, is a well-known oncogene implicated in various types of cancer, including pancreatic, colorectal, and lung cancers. The G12D mutation involves a substitution of glycine (G) with aspartic acid (D) at the 12th position of the K-Ras protein. This mutation leads to a constitutively active K-Ras protein that continuously signals for cell growth and division, bypassing normal regulatory mechanisms. As a result, cells harboring this mutation proliferate uncontrollably, contributing to tumorigenesis. The K-Ras (G12D) mutant is a critical target for cancer research and drug development, as targeting this mutation could potentially inhibit the aberrant signaling pathways and slow down or stop cancer progression. Despite the challenges in targeting mutant K-Ras proteins due to their high affinity for GTP/GDP and lack of suitable binding pockets, ongoing research is focused on developing specific inhibitors and novel therapeutic strategies to effectively target this mutation.
