Ku70 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry (Paraffin) analysis of Ku70 was performed in paraffin-embedded human cervical cancer tissue using Ku70 Polyclonal Antibody (Product # PA5-27538) at a dilution of 1:500.

Ku70 Antibody in Western Blot (WB) — Knockdown of Ku70 was achieved by transfecting HeLa cells with Ku70 specific siRNAs (Silencer® select Product # s5457, s5455). Western blot analysis (Fig. a) was performed using nuclear enriched cell extracts from the Ku70 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with Anti-Ku70 Polyclonal Antibody (Product # PA5-27538, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to Ku70.

Ku70 Antibody in Immunoprecipitation (IP) — PA5-27538 immunoprecipitates Ku70 protein in IP experiments. IP Sample: 1000 µg HeLa whole cell lysate/extract A. 40 µg HeLa whole cell lysate/extract B. Control with 2.5 µg of preimmune rabbit IgG C. Immunoprecipitation of Ku70 protein by 2.5 µg of Ku70 antibody (Product # PA5-27538) on 7.5% SDS-PAGE.

Ku70 Antibody — Antibody specificity was demonstrated by siRNA mediated knockdown of target protein. HeLa cells were transfected with Ku70 siRNA and decrease in signal was observed in Western Blot using Ku70 Polyclonal Antibody (Product # PA5-27538). {KD}

XRCC6 is a nuclear complex consisting of two subunits with molecular masses of approximately 70 and 80 kDa. The complex functions as a single-stranded DNA-dependent ATP-dependent helicase. The complex may be involved in the repair of nonhomologous DNA ends such as that required for double-strand break repair, transposition, and V(D)J recombination. High levels of autoantibodies to p70 and p80 have been found in some patients with systemic lupus erythematosus.
