5A6 specifically reacts with the 85 kD b-chain of human LRP/a2MW which is expressed in a restricted spectrum of cell types. A strong immunohistochemical reaction is seen in hepatocytes, tissue macrophages, subsets of neurones and astrocytes in the central nervous system, fibroblasts, smooth muscle cells, and monocyte-derived foam cells in atherosclerotic lesions in the arterial wall. The antibody can also be used for the characterization of a subset of myelo-monocytic subtypes of chronic and acute leukemia (CD 91). Neutralizing effects of 5A6 have also been reported (Basu et al., 2001).Basu S, Binder RJ, Ramalingam T, Srivastave PK: CD91 is a common receptor for heat shock proteins gp96, hsp90, hsp 70, and calreticulin. This gene encodes the major vault protein which is a lung resistance-related protein. Vaults are multi-subunit structures that may be involved in nucleo-cytoplasmic transport. This protein mediates drug resistance, perhaps via a transport process. It is widely distributed in normal tissues, and overexpressed in multidrug-resistant cancer cells. The protein overexpression is a potentially useful marker of clinical drug resistance. This gene produces two transcripts by using two alternative exon 2 sequences; however, the open reading frames are the same in both transcripts.
