Levodopa, also known as L-DOPA, is the most widely used drug for the symptomatic treatment of Parkinson's disease, serving as the gold standard for managing the disease's motor symptoms. It functions as a precursor to dopamine, which is deficient in patients with Parkinson's disease due to the degeneration of dopaminergic neurons in the substantia nigra. Once ingested, levodopa crosses the blood-brain barrier and is converted to dopamine, thereby alleviating symptoms such as bradykinesia, rigidity, and tremor. The pharmacokinetics and pharmacodynamics of levodopa have been extensively studied, with various formulations developed to optimize its absorption and reduce side effects. Despite its efficacy, long-term use of levodopa can lead to complications, including motor fluctuations and dyskinesias.
