Description: The monoclonal antibody AP20 recognizes the high molecular weight isoforms of microtubule-associated protein-2 (MAP2a and MAP2b) of human, rat, mouse, bovine, chicken, and xenopus. MAP2 belongs to a family of heat-stable phosphoproteins that are differentially expressed during brain development. MAP2 expression is localized to the cell body and dendrites of neurons and is involved in stabilizing microtubules by crosslinking with intermediate filaments. Expression of MAP2 is commonly used as a neural marker given that its expression becomes more pronounced with neuronal maturation.
Applications Reported: This AP20 antibody has been reported for use in immunohistochemical staining of frozen tissue sections, immunohistochemical staining of formalin-fixed paraffin embedded tissue sections, microscopy, and immunocytochemistry.
Applications Tested: This AP20 antibody has been tested by immunohistochemistry of formalin-fixed paraffin embedded human tissue using high or low antigen retrieval at less than or equal to 5 µg/mL. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
Filter Recommendation: When using this eFluor® 570 antibody conjugate, we recommend a filter that will capture the 570 emission wavelength (for example, Excitation 545/25, 565LP, Emission 605/70). A standard Alexa Fluor® 555 or TRITC filter is acceptable.
MAP2 (Microtubule Associated Protein 2) exists in two high molecular weight forms (MAP2a & MAP2b) and a low molecular weight form. The expression of MAP2 is developmentally regulated and its multiple forms arise by alternative splicing of a single gene. MAP2 is involved in microtubule assembly, which is an essential step in neurogenesis. The products of similar genes in rat and mouse are neuron-specific cytoskeletal proteins that are enriched in dendrites, implicating a role in determining and stabilizing dendritic shape during neuron development. MAP2 is useful in studies of neuron structure in normal and malignant brain tissue as well as in degenerative diseases such as Alzheimer's Disease (AD).
