MCM2 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of MCM2 was performed using 70% confluent log phase MCF-7 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. The cells were labeled with MCM2 Polyclonal Antibody (Product # PA5-78054) at 5µg/mL in 0.1% BSA, incubated at 4 degree Celsius overnight and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, Alexa Fluor® 488 (Product # A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing localization to nucleus. Panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

MCM2 Antibody in Western Blot (WB) — Western blot was performed using anti-MCM2 Polyclonal Antibody (Product # PA5-78054) and a 125 kDa band corresponding to MCM2 was observed across cell lines tested. Whole cell extracts (30 µg lysate) of MCF-7 (Lane 1), HeLa (Lane 2), A 549 (Lane 3), HCT-116 (Lane 4), SHSY-5Y (Lane 5) were electrophoresed using NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

MCM2 Antibody in Immunoprecipitation (IP) — Immunoprecipitation of MCM2 in HCT-116 whole cell extract. Samples were treated with MCM2 polyclonal antibody (Product # PA5-78054) using a dilution of 1:500. Results are as follows: A) 40 μg HCT-116 whole cell extract, B) Control with 4 μg of preimmune Rabbit IgG, C) Immunoprecipitation of MCM2 protein by 4 μg MCM2 antibody.

MCM2 Antibody — Antibody specificity was demonstrated by siRNA mediated knockdown of target protein. MCF-7 cells were transfected with MCM2 siRNA and reduction of signal was observed in Western Blot using MCM2 Polyclonal Antibody (Product # PA5-78054). {KD}

MCM2 (Mini chromosome maintenance protein 2) is involved in regulating DNA replication. MCM2 (also called CDCL1, mitotin and BM28), is a human nuclear protein that is crucial in the cell cycle, being involved in the onset of DNA replication and cell division. It is similar to members of the family of early S-phase proteins. MCM2 has been mapped to 3q21. From its localization, CDCL1 became a candidate for an oncogene affected by chromosomal breaks in acute myeloid leukemia (AML).
