Western blot analysis of MMP10 was performed by loading 30 µg of Mouse Pancreas (lane1) and Rat Pancreas (lane2) tissue lysates using Novex®NuPAGE®4-12 % Bis-Tris gel (Product # NP0321BOX), XCell SureLock Electrophoresis System (Product # EI0002), Novex® Sharp Pre-Stained Protein Standard (Product # LC5800), and iBlot® Dry Blotting System (Product # IB21001). MMP10 belongs to the matrix metalloproteinase family, and is generally secreted in its pro form, which is further modified by extracellular proteinases. As a metalloproteinase, MMP10 requires zinc and calcium as cofactors for activity. It is involved in the activation of pro-collagenase, catabolism of collagen, degradation of fibronectin, and tissue remodeling. In humans, the gene MMP10 located on chromosome 11.
