PACAP 27/38 refers to the two biologically active isoforms of pituitary adenylate cyclase-activating polypeptide (PACAP), a neuropeptide derived from the ADCYAP1 gene through proteolytic processing of a larger precursor protein. PACAP-38 is the predominant endogenous form, while PACAP-27 corresponds to the N-terminal 27 amino acids of PACAP-38. Both peptides function as potent signaling molecules that bind to PAC1, VPAC1, and VPAC2 receptors, activating adenylate cyclase and downstream cAMP-dependent pathways. PACAP-27/38 plays important roles in neurodevelopment, neurotransmission, neuroprotection, stress response, circadian regulation, and modulation of immune and endocrine functions. Due to their broad physiological activity and involvement in neurological, inflammatory, and metabolic disorders, PACAP isoforms are widely studied in both basic and translational research contexts.
