In Arabidopsis thaliana, PAL1-PAL4 encode closely related cytosolic phenylalanine ammonia-lyase isozymes that function as soluble enzymes in primary-to-secondary metabolism channeling. Each gene produces transcripts that can yield protein isoforms, and the proteins share the conserved ammonia-lyase fold and active-site architecture characteristic of plant PALs, supporting their role as homotetrameric enzymes in vivo. Collectively, PAL1-PAL4 catalyze the nonoxidative deamination of L-phenylalanine to trans-cinnamic acid, the committed entry step to the phenylpropanoid pathway, thereby supplying precursors for lignin and a wide range of specialized metabolites, including flavonoids and stress-induced phenolics. Genetic and expression analyses indicate substantial functional redundancy, with PAL1 and PAL2 particularly important for flavonoid/anthocyanin and seed coat tannin accumulation under environmental challenge, while the four-gene family as a whole contributes to developmentally and stress-regulated phenylpropanoid output (including wall-associated phenolics important for structural integrity and defense).
