PDR8 encodes a eukaryotic transcriptional regulator that localizes to the nucleus and functions as a zinc-cluster (Zn2Cys6) DNA-binding protein typical of fungal C6 transcription factors, with an N-terminal binuclear zinc-cluster DNA-binding domain and a larger C-terminal regulatory region that supports promoter binding and transcriptional activation. In Saccharomyces cerevisiae, Pdr8 modulates pleiotropic drug resistance by regulating expression of multiple detoxification and stress-response genes, including transporters from the ATP-binding cassette and major facilitator superfamily, thereby influencing cellular tolerance to xenobiotics and contributing to broader physiological traits such as growth on nonfermentable carbon sources and aspects of cell wall integrity; altered PDR8 dosage correspondingly changes resistance to specific compounds (e.g., increased resistance upon overexpression). In plants, PDR8 refers to an ATP-binding cassette transporter of the ABCG/PDR class that localizes to the plasma membrane and acts as an ATP-dependent efflux pump affecting the export of small molecules, with demonstrated roles in cadmium tolerance via extrusion of Cd species and in innate immunity by modulating the extent of pathogen-induced cell death, consistent with a broad substrate profile for defense- and stress-associated metabolites.
