The genes PPP2R2B, PPP2R2A, PPP2R2C, and PPP2R2D encode different regulatory subunits of protein phosphatase 2 (PP2A), a major serine/threonine phosphatase implicated in various cellular processes. PPP2R2B is located on chromosome 5q32 and has multiple alternatively spliced transcripts, playing a significant role in cancer prognosis and immune evasion (ScienceDirect). PPP2R2A, encoding the B55 alpha subunit, is frequently deleted in prostate adenocarcinomas, correlating with poor prognosis (PubMed). PPP2R2C, encoding the B55 gamma isoform, is shown to suppress glioma cell proliferation, acting as a tumor suppressor (ScienceDirect). PPP2R2D is involved in T-cell regulation and cancer cell proliferation, with overexpression linked to increased mTOR activity and impact on IL-2 production (PubMed). Collectively, these genes highlight the complexity and critical regulatory roles of PP2A subunits in cell signaling and disease pathology.
