Phospho-DAPK1 (Ser308) Antibody in Western Blot (WB) — Western blot analysis of Dapk1 Ser308ph was performed by loading whole cell lysate (in 1X SDS sample buffer with 2-ME) from 2 x 105 mouse embryonic fibroblasts. Samples were loaded onto a 4-12 % Bis-Tris polyacrylamide gel (Product # WG1402BOX). Proteins were transferred to nitrocellulose membrane by wet/tank transfer. Membrane was blocked in 5% milk/TBST and probed with a monoclonal anti-pDapk1 (S308) antibody (Product # MA1-24738) at a dilution of 1:1000 in 5% milk/TBST at 4°C overnight, followed by a secondary antibody HRP-anti-mouse at a dilution of 1:5000 at room temperature for 1 hour. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32209). The protein has a predicted MW of ~160 kDa.

DAPK1 (death-associated protein kinase 1) is a calcium/calmodulin-dependent serine/threonine kinase involved in multiple cellular signaling pathways that trigger cell survival, apoptosis, and authophagy. It regulates both type I apoptotic and type II autophagic cell death signal, depending on the cellular setting. Type I apoptotic cell death is caspase-dependent, type II authophagic cell death is caspase-independent. DAPK1 phosphorylates PIN1 resulting in inhibition of its catalytic activity, nuclear localization, and cellular function. It also phosphorylates TPM1, STX1A, PRKD1, BECN1, TSC2, RPS6, MYL9, and DAPK3.
