Phospho-VAV1 (Tyr160) Antibody in Immunoprecipitation (IP) — Vav1 protein immunoprecipitated from K562 cell extracts was resolved by SDS-PAGE on a 10% Tris-glycine gel and transferred to PVDF. Membranes were blocked with a 5% BSA-TBST buffer overnight at 4°C, then were incubated with 0.50 µg/mL Vav1 (pY160) antibody for two hours at room temperature in a 3% BSA-TBST buffer, following prior incubation with: no peptide (1), the non-phosphopeptide corresponding to the immunogen (2), a generic phosphotyrosine containing peptide (3), or, the phosphopeptide immunogen (4). After washing, membranes were incubated with goat F (ab')2 anti-rabbit IgG alkaline phosphatase (Product # ALI4405) and signals were detected using the Tropix WesternStar™ method. The data show that only the peptide corresponding to Vav1 (pY160) blocks the antibody signal, thereby demonstrating the specificity of the antibody.

Phosphorylated proteins were normalized to respective total proteins. Individual phospho-proteins, their respective total proteins, and beta-actin were presented from the same blots. The blots were first probed for phospho-proteins, followed by stripping and re-probing for respective total proteins and subsequently for beta-actin. This gene encodes the coiled-coil containing protein optineurin. Optineurin may play a role in normal-tension glaucoma and adult-onset primary open angle glaucoma. Optineurin interacts with adenovirus E3-14.7K protein and may utilize tumor necrosis factor-alpha or Fas-ligand pathways to mediate apoptosis, inflammation or vasoconstriction. Optineurin may also function in cellular morphogenesis and membrane trafficking, vesicle trafficking, and transcription activation through its interactions with the RAB8, huntingtin, and transcription factor IIIA proteins. Alternative splicing results in multiple transcript variants encoding the same protein.
