Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is colon carcinoma or brain tissue.
Pleiotrophin (PTN) is an 18 kDa secreted heparin and chondroitin-binding protein that is a regulatory cytokine critical to the early development of blood vessels and neurons. High PTN serum levels are associated with a variety of solid tumors. It was shown that patients with multiple myeloma (MM) also have elevated serum levels of this protein. The expression of this protein is very limited in normal adult tissues.
