SERCA1 ATPase Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of SERCA1 ATPase was performed using C2C12 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. The cells were labeled with SERCA1 ATPase Polyclonal Antibody (Product # PA5-78835) at 1:250 dilution in 0.1% BSA, incubated at 4 degree Celsius overnight and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing staining in endoplasmic reticulum. Panel e represents control cells with no primary antibody to assess background. SERCA1 ATPase Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry analysis of SERCA1 ATPase on paraffin-embedded mouse skeletal muscle tissue. Sample was incubated with SERCA1 ATPase polyclonal antibody (Product# PA5-78835).

SERCA1 ATPase Antibody in Western Blot (WB) — Western blot was performed using Anti-SERCA1 ATPase Polyclonal Antibody (Product # PA5-78835) and a 110 kDa bands corresponding to SERCA1 ATPase was observed in Mouse Skeletal Muscle, Rat Skeletal Muscle and not in Mouse Heart and Rat Heart which are reported negative for SERCA1 ATPase expression. Tissue extracts (30 µg lysate) of Mouse Skeletal Muscle (Lane 1), Rat Skeletal Muscle (Lane 2), Mouse Heart (Lane 3) and Rat Heart (Lane 4) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001) (Fig. a). The blot was probed with the primary antibody (0.1 µg/mL) and detected by chemiluminescence Goat Anti-Rabbit IgG Secondary Antibody, HRP conjugate (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). Relative expression of the SERCA1 ATPase in various tissues is shown in the histogram (Fig. b).

SERCA1 ATPase Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across tissues owing to their inherent genetic constitution. Relative expression of SERCA1 ATPase was observed in Mouse Skeletal Muscle, Rat Skeletal Muscle and not in Mouse Heart, Rat Heart using Anti- SERCA1 ATPase Polyclonal Antibody (Product # PA5-78835) in Western Blot. . {RE}

ATP dependent calcium pumps are responsible, in part, for the maintenance of low cytoplasmic free calcium concentrations. The ATP pumps that reside in intracellular organelles are encoded by a family of structurally related enzymes, termed the sarcoplasmic or endoplasmic reticulum calcium (SERCA) ATPases. The SERCA1 gene is exclusively expressed in type II (fast) skeletal muscle. The SERCA2 gene is subject to tissue dependent processing which is responsible for the generation of SERCA2a muscle-specific form expressed in type I (slow) skeletal, cardiac and smooth muscle and the SERCA2b isoform expressed in all cell types. The SERCA3 gene is not as well characterized and is found in non-muscle cells.
