Western blot analysis was performed on Nuclear extracts (30 µg lysate) of SH-SY5Y (Lane 1), HEL 92.1.7 (Lane 2), NTERA-2 (Lane 3) and U-2 OS (Lane 4). The blots were probed with Anti-SETMAR Rabbit Polyclonal Antibody (Product # 720343, 1-2 µg/mL) and detected by chemiluminescence using Goat anti-Rabbit IgG (Heavy Chain) Superclonal Secondary Antibody, HRP conjugate (Product # A27036, 0.4 µg/mL, 1:2500 dilution). SETMAR (SET domain and mariner transposase fusion gene) is a histone methyltransferase that methylates Lys-4 and Lys-36 of histone H3, creating specific tags for epigenetic transcriptional activation. It specifically mediates dimethylation of H3 Lys-36. SETMAR has sequence-specific DNA-binding activity, and recognizes the 19-mer core of the 5'-terminal inverted repeats (TIRs) of the Hsmar1 transposable element. SETMAT has DNA nicking activity, and in vivo end-joining activity, and may mediate genomic integration of foreign DNA.
