SOX2 Antibody — Fig. 1 Loss of Oct4 and maintenance of Sox2 expression during retinoic acid-induced differentiation. Oct4 expression begins to decline after 24-hour exposure to retinoic acid (1 µM), with the main transition to an Oct4- state occurring between 24 and 48 h of RA treatment. After 72 h, ~ 70% of the population retain an Oct4-Sox2 + phenotype (a, c). A schematic depicting the change in gene expression in ESC populations towards the predominant progenitor state during two differentiation protocols: LIF-withdrawal in serum-containing media and retinoic acid addition (b). SOX2 is an intronless gene encoding a member of the SRY-related HMG-box (SOX) family of transcription factors involved in the regulation of embryonic development and in the determination of cell fate. The product of the SOX2 gene is required for stem-cell maintenance in the central nervous system, and also regulates gene expression in the stomach. The SOX2 gene lies within an intron of another gene called SOX2 overlapping transcript (SOX2OT). Further, SOX2 protein may act as a transcriptional activator after forming a protein complex with other proteins. Mutations in the SOX2 gene have been associated with bilateral anophthalmia, a severe form of structural eye malformation, optic nerve hypoplasia and syndromic microphthalmia.
