Store lyophilized/reconstituted at -20°C; once reconstituted make aliquots to avoid repeated freeze-thaw cycles. Please, remember to spin tubes briefly prior to opening them to avoid any losses that might occur from lyophilized material adhering to the cap or sides of the tubes.

SYP121 encodes a plasma membrane-resident Qa-SNARE (syntaxin) that concentrates at the cell surface where secretory vesicles fuse and, in highly polarized contexts, is targeted to discrete plasma membrane domains (e.g., guard-cell and tip-growing cell regions). The protein is a single-pass membrane anchor with a C-terminal transmembrane helix, preceded by a cytosolic SNARE (Qa) motif that forms coiled-coil complexes with partner SNAREs, and an N-terminal regulatory region that includes an Habc-like folded domain and short N-terminal motifs important for partner binding and control of SNARE assembly. Functionally, SYP121 promotes post-Golgi exocytosis to the plasma membrane and helps define cargo delivery routes, thereby supporting growth and cell surface remodeling; it also couples membrane trafficking to transport activity by directly regulating and/or recycling key plasma membrane channels, including inward-rectifying K+ channels that drive guard-cell osmotic uptake during stomatal opening, and aquaporins that tune membrane water permeability. Beyond housekeeping secretion, SYP121 contributes to stimulus-responsive membrane fusion through regulated SNARE complex formation (including phosphorylation-dependent enhancement of pairing with R-SNARE partners) and participates in processes such as stomatal dynamics, polarized cell expansion, and defense-related cell wall fortification at attempted pathogen entry sites.
