SRK2D, SRK2E and SRK2I encode closely related, plant-specific SNF1-related protein kinase 2 (SnRK2) family members in Arabidopsis thaliana that function primarily in stress hormone signaling; their gene products are soluble kinases found predominantly in the nucleus in relevant developmental contexts and are activated by abscisic acid (ABA) and/or osmotic stress. Structurally, these proteins comprise an N-terminal serine/threonine protein kinase catalytic domain coupled to a C-terminal regulatory region that is required for stimulus-dependent activation and protein-protein interactions; for SRK2E, the C-terminal regulatory region can be functionally subdivided into domains that differentially contribute to ABA-dependent versus ABA-independent (osmotic stress) activation and can mediate interaction with the PP2C phosphatase ABI1. Functionally, SRK2D/E/I act as major positive regulators of ABA signaling during water-deficit responses and seed physiology: SRK2E is required for rapid stomatal closure under decreased humidity and for full induction of ABA-responsive genes, while combined loss of SRK2D, SRK2E and SRK2I causes strong ABA insensitivity and drought susceptibility with broadly impaired ABA- and dehydration-induced transcription, consistent with their redundant control of downstream transcription factor phosphorylation and gene expression programs.
