TDP-43 Antibody in Western Blot (WB) — Western blot was performed using Anti-TDP-43 Polyclonal Antibody(Product # PA5-19064) and a 44kDa band corresponding to TDP-43 was observed across cell lines tested. Whole Cell Extract-WCL (30 µg lysate) of HeLa (Lane 1), MCF7 (Lane 2), Caco-2 (Lane 3), K-562 (Lane 4), Neuro-2a (Lane 5) and NIH/3T3 (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.3 µg/mL) and detected by chemiluminescence with Rabbit anti-Goat IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27014, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

TDP-43 Antibody — Antibody specificity was demonstrated by siRNA mediated KD of target protein. HeLa cells were transfected with TDP-43 siRNA and decrease in signal intensity was observed in Western Blot application using Anti-TDP-43 Polyclonal Antibody (Product # PA5-19064). {KD}

This antibody is predicted to react with bovine, canine, porcine and rat based on sequence homology.

TDP43 was first identified as a novel cellular protein that binds to HIV-1 virus TAR DNA sequence motifs and acts a transcriptional repressor to the HIV-1 LTR. Later experiments revealed that TDP43 also regulates the splicing of exon 9 of the cystic fibrosis transmembrane conductance regular (CFTR), most likely through the association with the UG repeats at the 3"e; splice site. A hyperphosphorylated, ubiquitinated, and cleaved form of TDP43 known as pathologic TDP43 is the major disease protein in ubiquitin-positive, tau-, and alpha-synuclein-negative frontotemporal dementia (FLTD-U). TDP43 is not related to TRBP1, and RNA binding protein that binds HIV-1 TAR RNA sequences.
