TNXB Antibody in Immunocytochemistry (ICC/IF) — Immunocytochemistry analysis of TNXB using anti-TNXB antibody (Product # PA5-80173). TNXB was detected in a section of MCF7 cells. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent for 15 mins. The cells were blocked with 10% goat serum and then incubated with 2μg/mL rabbit anti-TNXB antibody (Product # PA5-80173) overnight at 4°C. DyLight®594 Conjugated Goat Anti-Rabbit IgG was used as secondary antibody at 1:100 dilution and incubated for 30 minutes at 37°C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.

TNXB Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry analysis of TNXB on paraffin-embedded human placenta tissue. Sample was incubated with TNXB polyclonal antibody (Product# PA5-80173).

TNXB Antibody in Western Blot (WB) — Western blot analysis of TNXB in Lane 1: MCF- 7 whole cell lysate, Lane 2: SW620 whole cell lysate, Lane 3: HeLa whole cell lysate using 40-50 µg per well. Sample was incubated with TNXB (Product # PA5-80173) at a dilution of 0.5 µg/mL.

TNXB Antibody in Flow Cytometry (Flow) — Flow Cytometry of TNXB in HeLa cells (blue line), isotype control rabbit IgG (green line) and unlabeled (red line). Samples were blocked with 10% goat serum, incubated with TNXB Polyclonal Antibody (Product # PA5-80173) at a dilution of 1 μg (per 1x10^6 cells), followed by DyLight®488 conjugated goat anti-rabbit IgG (for 30 minutes at 20°C) using 5-10 μg (per 1x10^6 cells) dilution.

Tenascin-X (TNXB) is a extracellular matrix glycoprotein predominantly located in the outer reticular lamina of the basement membrane (BM). It interacts with many other ECM proteins and it accelerates collagen fibrillogenesisin vitro. TNXB plays a role in interactions between cell and ECM, antiadhesive effect, inhibiting cell migration, and in maintaining homeostasis of the extracellular matrix. Deficiency of TNXB has been associated with the connective tissue disorder Ehlers-Danlos syndrome.
