The ZCCHC4 gene encodes zinc finger CCHC domain-containing protein 4, a member of the zinc finger CCHC-type (ZCCHC) superfamily characterized by a consensus sequence C-X2-C-X4-H-X4-C. This gene plays a vital role in binding single-stranded nucleic acids, particularly RNAs, and is involved in multiple cellular processes. Notably, ZCCHC4 has been implicated in cancer, with aberrantly high expression linked to poor prognosis and chemoresistance in various cancers, including hepatocellular carcinoma, pancreatic cancer, colon cancer, and esophageal cancer. The protein exhibits RNA-modifying capabilities, influencing cell growth and survival. Recent studies have identified the structural mechanisms through which ZCCHC4 interacts with RNA substrates, offering insights into its role in regulating cancer progression and therapeutic resistance. In esophageal cancer, downregulation of ZCCHC4 may enhance sensitivity to chemotherapeutic agents like cisplatin, indicating its potential as a target for cancer treatment strategies.
