Pro-Kallikrein 5 (KLK5) is a member of the kallikrein-related peptidase family, which comprises serine proteases involved in various physiological processes. KLK5 is encoded by the KLK5 gene located on chromosome 19 in humans. This enzyme is initially synthesized as an inactive zymogen, pro-Kallikrein 5, and is activated through proteolytic cleavage. KLK5 plays a critical role in skin desquamation, the process of shedding dead skin cells, by degrading intercellular adhesive proteins in the epidermis. It is also implicated in the regulation of inflammation and immune responses. Abnormal expression or activity of KLK5 has been linked to skin disorders such as Netherton syndrome and atopic dermatitis, as well as certain cancers. Due to its involvement in these conditions, KLK5 is being studied as a potential biomarker and therapeutic target for disease diagnosis and treatment. Understanding the regulation and function of KLK5 can provide insights into skin homeostasis and the pathogenesis of related diseases.
