The Bovine Cortisol ELISA quantitates Cortisol in serum and plasma.
原理方法
The Bovine Cortisol solid-phase competitive ELISA (enzyme-linked immunosorbent assay) is designed to measure the concentration of Cortisol in biological samples. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody.
Bovine Cortisol in samples (or standards) competes with a fixed amount of Cortisol immobilized on the solid phase for binding to the HRP-linked detection antibody specific to Cortisol. Excess conjugate and unbound sample or standard are washed from the plate.
A substrate solution is added that reacts with the enzyme-antibody complex to produce a measurable signal. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The intensity of this signal, measured spectrophotometrically at a wavelength of 450 ± 2 nm, is inversely proportional to the concentration of Cortisol present in the original specimen.

Cortisol is the most potent glucocorticoid produced by the human adrenal. It is synthesized from cholesterol and its production is stimulated by pituitary adrenocorticotropic hormone (ACTH) which is regulated by corticotropin releasing factor (CRF). ACTH and CRF secretions are inhibited by high cortisol levels in a negative feedback loop. In plasma a majority of cortisol is bound with high affinity to corticosteroid binding globulin (CBG or transcotin). Cortisol acts through specific intracellular receptors and affects numerous physiologic systems including immune function, glucose counter regulation, vascular tone, and bone metabolism.
