The Human FXN (Frataxin) ELISA quantitates FXN in serum and plasma.
原理方法
The Human Frataxin (FXN) solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the concentration of Frataxin (FXN) in biological samples. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody.
The sandwich complex is formed by the addition of a second (HRP-linked) detection antibody specific to Frataxin (FXN). Excess reagents are washed from the plate.
A substrate solution is added that reacts with the enzyme-antibody-target complex to produce a measurable signal. The enzyme-substrate reaction is terminated by the addition of stop solution. The intensity of the signal, measured spectrophotometrically at 450 ± 2 nm, is directly proportional to the concentration of Frataxin (FXN) present in the original specimen.

FXN is a mitochondrial protein which belongs to the FRATAXIN family. The protein functions in regulating mitochondrial iron transport and respiration. The expansion of intronic trinucleotide repeat GAA results in Friedreich ataxia.
