The Human CXCL1 (Growth Regulated Oncogene Alpha) ELISA quantitates CXCL1 in serum and plasma.
Principle of the method
The Human Growth Regulated Oncogene Alpha (CXCL1) solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the concentration of Growth Regulated Oncogene Alpha (CXCL1) in biological samples. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody.
The sandwich complex is formed by the addition of a second (HRP-linked) detection antibody specific to Growth Regulated Oncogene Alpha (CXCL1). Excess reagents are washed from the plate.
A substrate solution is added that reacts with the enzyme-antibody-target complex to produce a measurable signal. The enzyme-substrate reaction is terminated by the addition of stop solution. The intensity of the signal, measured spectrophotometrically at 450 ± 2 nm, is directly proportional to the concentration of Growth Regulated Oncogene Alpha (CXCL1) present in the original specimen.

CXCL1, CXCL2, and CXCL3, also known respectively as GRO alpha, GRO beta (MIP-2 alpha) and GRO gamma (MIP-2 beta), are members of the CXC subfamily of chemokines. Mature CXCL1/2/3 proteins bind with high affinity to the IL-8 receptor type B and are potent neutrophil attractants and activators. In vitro, the processed forms GRO-alpha(4-73), GRO-alpha(5-73) and GRO-alpha(6-73) show a 30-fold higher chemotactic activity.
