Cryo-electron tomography (cryo-ET) allows visualization of organelles and cellular structures in their near-native state at resolutions far beyond light microscopy. Recent advancements have made cryo-ET workflows more accessible, enabling even non-expert users to produce high-quality cryo-lamellae for statistically relevant data. Innovations such as the vacuum capsule, which links the Thermo Scientific Arctis Cryo-Plasma-FIB with the TEM, minimize ice contamination during transfer and improve data quality and throughput. Combined with the Thermo Scientific Krios 5 Cryo-TEM, these developments ensure efficient transfer and processing of large numbers of lamellae. User-friendly software further simplifies lamella preparation, making cryo-ET more accessible to cell biologists.

However, this process is challenging due to the inherent low contrast of cryo imaging and the complexity of the internal cellular environment. This talk explores the combination of Volume Electron Microscopy (vEM) and Cryo-Electron Tomography (cryo-ET), enabling visualization of cellular landscapes in their native state. vEM techniques, like Focused Ion Beam Scanning Electron Microscopy (FIB-SEM), provide 3D reconstructions with nanometer precision, offering essential context to ultrastructural details. Cryo-ET complements this by imaging biomolecules in near-native conditions, providing molecular-level detail. Recent advancements have automated and scaled cryo-ET workflows, delivering the statistical validation required in cell biology.

Cell and gene therapies have emerged as powerful tools for treating cancer, autoimmune disorders, and genetic diseases, while iPSCs have drawn unique interest for the development of clinically manufacturing CAR-NK and CAR-T cells. These approaches often rely on the ability to genetically modify immune or stem cells ex vivo, requiring robust, non-viral delivery platforms that can maintain efficiency and scalability from early-stage research through clinical manufacturing. Electroporation is key technology in this space, allowing delivery of DNA, RNA, or protein payloads into hard-to-transfect cell types without the complexity and limitations of viral systems. This talk will describe the entire workflow from CAR-iPSC engineering optimization techniques to obtaining final product of functional tumor-specific (iPSC)-derived natural killer (CAR-iNK) cells with enhanced cytotoxicity and precision.
