Cyclin-dependent kinase 6 (CDK6) and Cyclin D1 are critical components of the cell cycle regulation, particularly in the transition from the G1 to the S phase. CDK6 is a protein kinase that requires association with cyclins, such as Cyclin D1, to become enzymatically active. This partnership is essential for phosphorylating target substrates that drive cell cycle progression. CDK6 and Cyclin D1 together form complexes that regulate the retinoblastoma protein (Rb) pathway, which is a key mechanism controlling cell proliferation. The expression and activation of CDK6/Cyclin D1 complexes are tightly regulated, and imbalances can contribute to oncogenesis. Structural studies have shown that CDK6 has a unique C-terminal region crucial for its nuclear transport and localization, which is essential for its function. Cyclin D1's dynamics-its expression, accumulation, and degradation-are pivotal in its regulatory roles, especially its interactions with CDK4 and CDK6. In cancer, genetic alterations affecting CDK6 and Cyclin D1 can lead to uncontrolled cell division, emphasizing their significance in cancer progression and as potential therapeutic targets.
