The mature form of mouse KC was expressed in Pichia pastoris, X-33. GenBank Accession # NP_032202, amino acids 20 - 96. For protein verification, the expressed protein was N-terminal sequenced and begins with the following 10 amino acids: RLATGAPIAN. Purity is >95% by SDS-PAGE analyses.
Activity: The biological activity of recombinant mouse KC was measured by its ability to induce neutrophil migration in vitro. The ED50 (or 1 unit of activity) for this effect in our laboratories is typically 3 pg/mL.
Use: For in vitro applications. A dose-response assay should be performed to determine the optimal concentration for specific experiments.
Reconstitution and Dilution: Store lyophilized material at 4°C. Reconstitute in 0.5 mL of distilled or deionized water. Dilute in a buffer-containing carrier protein, aliquot, and store at or below -70°C, avoiding multiple freeze-thaw cycles. PBS with 1% BSA or a neutral media containing <10% FCS are suitable buffers.
Endotoxin level is <0.01 EU/ug.
CXCL1, CXCL2, and CXCL3, also known respectively as GRO alpha, GRO beta (MIP-2 alpha) and GRO gamma (MIP-2 beta), are members of the CXC subfamily of chemokines. Mature CXCL1/2/3 proteins bind with high affinity to the IL-8 receptor type B and are potent neutrophil attractants and activators. In vitro, the processed forms GRO-alpha(4-73), GRO-alpha(5-73) and GRO-alpha(6-73) show a 30-fold higher chemotactic activity.
