The Human CD147 (Extracellular Matrix Metalloproteinase Inducer) ELISA quantitates CD147 in serum and plasma.
原理方法
The Human Extracellular Matrix Metalloproteinase Inducer (CD147) solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the concentration of Extracellular Matrix Metalloproteinase Inducer (CD147) in biological samples. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody.
The sandwich complex is formed by the addition of a second (HRP-linked) detection antibody specific to Extracellular Matrix Metalloproteinase Inducer (CD147). Excess reagents are washed from the plate.
A substrate solution is added that reacts with the enzyme-antibody-target complex to produce a measurable signal. The enzyme-substrate reaction is terminated by the addition of stop solution. The intensity of the signal, measured spectrophotometrically at 450 ± 2 nm, is directly proportional to the concentration of Extracellular Matrix Metalloproteinase Inducer (CD147) present in the original specimen.

CD147 (basigin, neurothelin, OX-47, 5A11, CE9, M6, EMMPRIN, extracellular matrix metalloproteinase inducer, TCSF, tumour cell-derived collagenase-stimulatory factor) is a 50-60 kDa glycoprotein with multiple glycosylated forms. CD147 is a plasma membrane protein that is important in spermatogenesis, embryo implantation, neural network formation, and tumor progression. CD147 is also a member of the immunoglobulin superfamily. The highest level of CD147 expression is on metabolically active cells, such as lymphoblasts, inflammatory cells, brown adipocytes and malignant tumor cells. CD147 has multiple functions, including facilitating of cell surface expression of monocarboxylate transporter proteins and extracellular matrix metalloproteinases, regulation of integrin functions, it plays roles in cell development and activation, fetal development or retinal function.
