The Human HO1 (Heme Oxygenase 1) ELISA quantitates HO1 in serum and plasma.
Principle of the method
The Human Heme Oxygenase 1 (HO1) solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the concentration of Heme Oxygenase 1 (HO1) in biological samples. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody.
The sandwich complex is formed by the addition of a second (HRP-linked) detection antibody specific to Heme Oxygenase 1 (HO1). Excess reagents are washed from the plate.
A substrate solution is added that reacts with the enzyme-antibody-target complex to produce a measurable signal. The enzyme-substrate reaction is terminated by the addition of stop solution. The intensity of the signal, measured spectrophotometrically at 450 ± 2 nm, is directly proportional to the concentration of Heme Oxygenase 1 (HO1) present in the original specimen.

Heme oxygenase (HO) is a microsomal enzyme that catalyzes the oxidation of heme to the antioxidant molecules, biliverdin and carbon monoxide. HO consists of two homologous isozymes, an inducible HO-1 and a constitutively expressed HO-2. HO-1 is induced by a wide variety of stimuli including conditions of oxidative stress, inflammatory agents, transforming growth factor beta and heat shock. The increase in expression of HO-1 is thought to be a cellular defense mechanism against oxidative stress since elevated HO could eventually generate more bilirubin, an anti-oxidant.
