The Human INS (Insulin) ELISA quantitates INS in serum and plasma.
原理方法
The Human Insulin (INS) solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the concentration of Insulin (INS) in biological samples. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody.
The sandwich complex is formed by the addition of a second (HRP-linked) detection antibody specific to Insulin (INS). Excess reagents are washed from the plate.
A substrate solution is added that reacts with the enzyme-antibody-target complex to produce a measurable signal. The enzyme-substrate reaction is terminated by the addition of stop solution. The intensity of the signal, measured spectrophotometrically at 450 ± 2 nm, is directly proportional to the concentration of Insulin (INS) present in the original specimen.

Insulin is a peptide hormone secreted by beta cells of the pancreatic islets. It regulates carbohydrate, protein and lipid metabolism by enhancing membrane transport of glucose, amino acids, and certain ions. It also promotes glycogen storage, formation of triglycerides and synthesis of proteins and nucleic acids. Insulin is 51-amino acid polypeptide product produced from a precursor peptide, proinsulin. Proinsulin is post-translationally cleaved into two chains (peptide A and peptide B) that are covalently linked via two disulfide bonds and one molecule of C-peptide. Insulin deficiency results in diabetes mellitus, one of the leading causes of morbidity and mortality in the general population. Insulin is also present in tumors of b-cell origin such as insulinoma.
