The Human PCSK9 (Proprotein Convertase Subtilisin/Kexin Type 9) ELISA quantitates PCSK9 in serum and plasma.
Principle of the method
The Human Proprotein Convertase Subtilisin/Kexin Type 9 (PCSK9) solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the concentration of Proprotein Convertase Subtilisin/Kexin Type 9 (PCSK9) in biological samples. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody.
The sandwich complex is formed by the addition of a second (HRP-linked) detection antibody specific to Proprotein Convertase Subtilisin/Kexin Type 9 (PCSK9). Excess reagents are washed from the plate.
A substrate solution is added that reacts with the enzyme-antibody-target complex to produce a measurable signal. The enzyme-substrate reaction is terminated by the addition of stop solution. The intensity of the signal, measured spectrophotometrically at 450 ± 2 nm, is directly proportional to the concentration of Proprotein Convertase Subtilisin/Kexin Type 9 (PCSK9) present in the original specimen.

PCSK9 is a proprotein convertase belonging to the proteinase K subfamily of the secretory subtilase family. This protein is synthesized as a soluble zymogen that undergoes autocatalytic intramolecular processing in the endoplasmic reticulum. The protein may function as a proprotein convertase. The protein plays a role in cholesterol homeostasis and may have a role in the differentiation of cortical neurons.
