The Human sTREM-1 (Triggering receptor expressed on myeloid cells 1) ELISA quantitates sTREM-1 in serum and plasma.
原理方法
The Human Triggering receptor expressed on myeloid cells 1 (sTREM-1) solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the concentration of Triggering receptor expressed on myeloid cells 1 (sTREM-1) in biological samples. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody.
The sandwich complex is formed by the addition of a second (HRP-linked) detection antibody specific to Triggering receptor expressed on myeloid cells 1 (sTREM-1). Excess reagents are washed from the plate.
A substrate solution is added that reacts with the enzyme-antibody-target complex to produce a measurable signal. The enzyme-substrate reaction is terminated by the addition of stop solution. The intensity of the signal, measured spectrophotometrically at 450 ± 2 nm, is directly proportional to the concentration of Triggering receptor expressed on myeloid cells 1 (sTREM-1) present in the original specimen.

TREM1 (triggering receptor expressed on myeloid cells 1) is a receptor belonging to the Ig superfamily that amplifies neutrophil and monocyte-mediated inflammatory responses triggered by bacterial and fungal infections by stimulating release of pro-inflammatory chemokines and cytokines, as well as increased surface expression of cell activation markers. It is thought that TREM1 other related TREM proteins such as TREM2 act to modulate the inflammatory response. TREM1 has also been linked to inflammatory bowel disease resulting in enhanced production of TNF, IL-6, IL-8, and MCP-1.
