The Human COMP (Cartilage Oligomeric Matrix Protein) ELISA quantitates COMP in serum and plasma.
原理方法
The Human Cartilage Oligomeric Matrix Protein (COMP) solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the concentration of Cartilage Oligomeric Matrix Protein (COMP) in biological samples. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody.
The sandwich complex is formed by the addition of a second (HRP-linked) detection antibody specific to Cartilage Oligomeric Matrix Protein (COMP). Excess reagents are washed from the plate.
A substrate solution is added that reacts with the enzyme-antibody-target complex to produce a measurable signal. The enzyme-substrate reaction is terminated by the addition of stop solution. The intensity of the signal, measured spectrophotometrically at 450 ± 2 nm, is directly proportional to the concentration of Cartilage Oligomeric Matrix Protein (COMP) present in the original specimen.

COMP is a noncollagenous extracellular matrix protein. It consists of five identical glycoprotein subunits, each with EGF like and calcium binding (thrombospondin like) domains. Oligomerization results from formation of a five stranded coiled coil and disulfides. Binding to other ECM proteins such as collagen appears to depend on divalent cations. Mutations can cause the osteochondrodysplasias pseudochondroplasia (PSACH) and multiple epiphyseal dysplasia (MED).
