The Human VCAM-1/CD106 (Vascular Cell Adhesion Molecule 1) ELISA quantitates VCAM-1 in serum and plasma.
Principle of the method
The Human Vascular Cell Adhesion Molecule 1 (VCAM-1/CD106) solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the concentration of Vascular Cell Adhesion Molecule 1 (VCAM-1/CD106) in biological samples. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody.
The sandwich complex is formed by the addition of a second (HRP-linked) detection antibody specific to Vascular Cell Adhesion Molecule 1 (VCAM-1/CD106). Excess reagents are washed from the plate.
A substrate solution is added that reacts with the enzyme-antibody-target complex to produce a measurable signal. The enzyme-substrate reaction is terminated by the addition of stop solution. The intensity of the signal, measured spectrophotometrically at 450 ± 2 nm, is directly proportional to the concentration of Vascular Cell Adhesion Molecule 1 (VCAM-1/CD106) present in the original specimen.

VCAM-1 is a cell surface sialoglycoprotein expressed by cytokine activated endothelium. The protein has a number of functions including the regulation of leukocyte migration, leukocyte-endothelial cell adhesion and signal transduction and may play a role in a number of inflammatory diseases.
