IP3 Competitive ELISA Kit quantifies IP3 in serum and plasma.
The IP3 Competitive ELISA research-use-only kit is designed to quantitatively measure IP3. A IP3 standard is provided to generate a standard curve for the assay and all samples are read off a user-generated standard curve. Standards or diluted samples are pipetted into a coated microtiter plate and Biotinylated Detection Ab specific to IP3 is added. Excess conjugate and unbound sample or standard are washed from the plate, and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color change is measured on a microplate reader. The concentration of IP3 in the samples is then determined by comparing the OD of the samples to the standard curve..

Inositol 1,4,5-trisphosphate (IP3) is a crucial intracellular signaling molecule that plays an essential role in various cellular processes by regulating calcium release from the endoplasmic reticulum. IP3 is generated through the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) by the enzyme phospholipase C (PLC) following receptor activation. The released IP3 binds to its receptors, inositol 1,4,5-trisphosphate receptors (ITPRs), which are located on the endoplasmic reticulum membrane. This binding triggers the release of Ca2+ ions into the cytoplasm, leading to numerous downstream effects, including muscle contraction, secretion, metabolism, and cell growth. ITPRs are encoded by different genes leading to multiple isoforms (ITPR1, ITPR2, and ITPR3), each showing distinct tissue distribution and regulatory properties.
