The Mouse LAB7-1 (B-Lymphocyte Activation Antigen B7-1) ELISA quantitates LAB7-1 in serum and plasma.
原理方法
The Mouse B-Lymphocyte Activation Antigen B7-1 (LAB7-1) solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the concentration of B-Lymphocyte Activation Antigen B7-1 (LAB7-1) in biological samples. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody.
The sandwich complex is formed by the addition of a second (HRP-linked) detection antibody specific to B-Lymphocyte Activation Antigen B7-1 (LAB7-1). Excess reagents are washed from the plate.
A substrate solution is added that reacts with the enzyme-antibody-target complex to produce a measurable signal. The enzyme-substrate reaction is terminated by the addition of stop solution. The intensity of the signal, measured spectrophotometrically at 450 ± 2 nm, is directly proportional to the concentration of B-Lymphocyte Activation Antigen B7-1 (LAB7-1) present in the original specimen.

CD80 (B7-1) and CD86 (B7-2) are ligands of T cell critical costimulatory molecule CD28, and of an inhibitory receptor CTLA-4 (CD152). Both B7 molecules are expressed on professional antigen-presenting cells and are essential for T cell activation, and both molecules can also substitute for each other in this process. CD80 is rapidly induced on the surface of in vitro activated B cells, with Epstein Barr Virus (EBV) transformed B cell lines, with Burkitts lymphoma cell lines, with freshly isolated follicular B lymphoma cells, T cells, and monocytes. The B-lymphocyte activation antigen B7-1 provides regulatory signals for T lymphocytes as a consequence of binding to the CD28 and CTLA4 ligands of T cells. Diseases associated with CD80 dysfunction include gallbladder squamous cell carcinoma and myocarditis.
