Representative data for AKT Multispecies InstantOne™ ELISA. MCF7 cells were treated with 10 µM wortmannin for 60 min (-) or stimulated with 10 ng/mL insulin for 10 min (+). Cell lysates were analyzed for levels of Total AKT 1 (left), phospho-AKT 1/2/3 (Ser473) (center), and phospho AKT 1/2/3 (Thr308) (right) using the AKT Activation InstantOne™ ELISA.

The InstantOne™ ELISA is specifically engineered for accurate measurement of human, mouse, and hamster total AKT 1 and phosphorylated AKT 1/2/3 (Thr308 and Ser473) in cell lysates. Detection of phosphorylated rat AKT 1/2/3 (Thr308 and Ser473) and total rat AKT 1 is also expected. The InstantOne™ ELISA kit allows for fast analysis of samples in approximately one hour. All reagents used in a traditional sandwich ELISA are added in solution to a plate followed by a wash step and detection with the TMB colorimetric substrate.

AKT also known as protein kinase B (PKB) or RAS-alpha, is an ubiquitous serine/threonine kinase that plays an important role in diverse biological responses such as regulation of metabolism, cell survival and growth by phosphorylating multiple proteins. This protein kinase is activated by insulin, PI3K, IGF1 and various other growth and survival factors. Akt promotes cell survival by inhibiting apoptosis through phosphorylation and inactivation of several targets, including forkhead transcription factors, and caspase-9. The AKT pathway is a major target for cancer drug discovery.
