Pepsinogen is the inactive precursor form of Pepsin, one of the main proteolytic enzymes secreted by the gastric mucosa. Pepsinogen is synthesized in the gastric mucosa by chief cells. After secretion, pepsinogen is activated in the presence of HCl, which creates an acidic environment in the stomach, allowing pepsinogen to unfold and cleave itself in an autocatalytic fashion. Pepsin consists of a single polypeptide chain and arises from its precursor, pepsinogen, by removal of a 41 amino acid segment from the N-terminus.
