关于 Dynabeads™ 人 T 活化剂 CD3/CD28 Dynabeads™ 人 T 活化剂 CD3/CD28 提供了一种简单的 T 细胞活化与扩增方法,无需饲养层细胞(抗原递呈细胞)或抗原。直径均匀为 4.5 µm 的惰性超顺磁珠的大小与抗原呈递细胞相似,同时共价偶联抗 CD3 和抗 CD28 抗体。这两种抗体提供了主要和共刺激信号,优化用于高效 T 细胞活化和扩增。可使用重组 IL-2 刺激 T 细胞群扩增,活化或扩增后,可使用 DynaMag™ 磁力架轻松地去除磁珠。对于从 T 细胞克隆或 T 细胞系扩增抗原特异性 T 细胞,我们建议使用 Dynabeads™ 人 T 活化剂 CD3/CD28/CD137。
起始样品 以来自全血或血沉棕黄层的单核细胞 (MNC)/外周血单核细胞 (PBMC) 或 T 细胞亚群(包括 CD3+ T 细胞、CD4+ T 细胞或 CD8+ T 细胞或调节性 T 细胞)开始。
下游应用 活化 T 细胞可在活化后不久进行分析(用于转染/转导或研究 TCR 信号转导、蛋白质组学、基因表达等)。培养基中可留存 T 细胞以分化为辅助性 T 细胞亚群或扩增多克隆/抗原特异性 T 细胞。
