Grace’s昆虫培养基中包含了哪些辅剂?为什么它们以单管形式提供?

补充培养基简称为TNM-FH。它包含以下成份(每500 mL):

TC酵母粉:1.66 g,乳清蛋白水解物1.66 g,L-谷氨酰胺:0.3 g
这些添加剂能够帮助增加培养基的稳定性和延长保质期。请注意这些添加剂未经消毒,须经过滤灭菌(通过0.2 µm滤器)后方可加入。
TNM-FH未加入热灭活的10% FBS之前,不视为完全培养基。血清会提供额外的养分,同时也能够保护悬浮培养条件(即旋转器和振荡培养瓶)下的细胞免受流体压力的影响。

For how long can I store Grace's Insect Medium, unsupplemented at room temperature?

We ship this product under “ambient” conditions throughout the year. We have performed shipping studies that shown components to be stable for at least 7 days at ambient temperatures. The important factor is to ensure that the media is not exposed to light at the warmer temperatures as there are vitamins and amino acids that will breakdown with light exposure.

What additional supplements are required in Grace's Insect Cell Culture Medium, Unsupplemented to make a "complete" formulation that will support insect cell growth?

Grace's Insect Cell Culture Medium, Unsupplemented requires addition of 3.33 g/L of lactalbumin hydrolysate and 3.33 g/L of yeastolate for the “base” growth medium. For the “complete” growth medium that will support insect cell growth, 10% fetal bovine serum must also be added. This “complete” growth medium is also referred to as TNM-FH.
