The substrate solution should remain colorless upon reaction with Horseradish peroxidase (HRP), if the HRP concentration is within the appropriate range. Extremely high concentrations of HRP can cause the substrate color to turn yellow/orange/brown. Alternatively, the color change of the substrate may be due to the presence of some other contaminant.
