With a 433A synthesizer, when switching from HBTU/DMF to HATU-DMF, do I need to change flow tests, specifications for the cycles or anything else?

No changes in the software or the delivery times are needed to replace the HBTU (HBTU Activation Kit, P/N 4425642) with HATU in DMF (kit part number 4317033). This is also true for any of the other "uronyl" activators (HCTU, TBTU, etc.). Since any excess of any of these activators, relative to the moles of amino acid being activated, results in resin-site termination, please continue to test carefully for delivery time. Use flow test "D" or #13, "bottle #5 to a cartridge", to determine the correct number of seconds to deliver the acceptable mass of reagent. Once this time, in seconds, is determined for your 433A, then modify the delivery times as needed in the SynthAssist chemistries (activation modules).

If I am sensitized to DCC, will I also have problems with HBTU in peptide synthesis? If I am sensitized to HATU, will I also have problems with HBTU?

Sensitization (an allergic response either on the skin or in the respiratory system) is individual-specific as well as chemical-specific. Just because one is allergic to DCC or HATU, does not mean that one is automatically allergic to HBTU or vice versa. All of these substances have been known to induce an allergic response to certain individuals or in animal studies.
