During the extraction process, the media inside the spin column removes PCR inhibitors from the sample, allowing the bacteria to pass through into the sample collection tube. The pellet is resuspended, then transferred (to avoid carryover, which inhibits PCR) to a new tube containing zirconium beads for mechanical lysis.
