Yes, if properly stored and within the recommended library sizes, the libraries can still be sequenced using the Ion PGM or Proton, and S5/S5XL systems. If the libraries were barcoded with the Ion DNA Barcoding 1-16 Kit (Cat. No. 4468654), please select ‘IonSet1' from the Barcode dropdown menu to trigger the proper barcode identification and splitting during sequencing run analysis on the Torrent Server.

The Ion DNA Barcoding 1-16 Kit contains different barcode sequences and the method by which the barcode sequences were added to the library is different compared to the current Ion Xpress Barcode Adapter kits. The current kit adds the barcode sequences as part of the adapter ligation process, while the old kit added the barcodes via PCR during the final library amplification.
