I tested the Mammalian beta-Galactosidase Assay Kit and did not use the Stop Solution. I performed three absorbance readings and the values were different, why is this?

Using the Assay Stop Solution is optional. However, if the Stop Solution is not used, the absorbance values from the plate will change over time. This means that the absorbance needs to be read immediately after the 30 min incubation.

How do I prepare cells for the Mammalian beta-Galactosidase Assay Kit?

Thermo Scientific M-PER Mammalian Protein Extraction Reagent (Cat. No.78503) may be used to prepare lysate from washed adherent or pelleted cells, and then a portion of the lysate can be used to assay for beta-galactosidase activity.
